Researchers have identified several proteins involved in the development of melanoma. One of the proteins – PDIA6 – was found very important to encourage malignancy. Experiments with mice show that melanoma cells with reduced levels of PDIA6 experience impaired ability to metastasize to the lungs. These findings can cause new therapeutic strategies that prevent the spread of melanoma from one part of the body to another.#RNAfishing #reveals #drivermelanoma #malignancymetastasisResearchers at the Center for Genomic Regulation have identified several proteins involved in the development of melanoma, the brightest form of skin cancer. One of the proteins – PDIA6 – was found very important to encourage malignancy. Experiments with mice show that melanoma cells with reduced levels of PDIA6 experience impaired ability to metastasize to the lungs. The researchers found that PDIA6 promoted melanoma malignancy by binding RNA molecules in tumor cells. Research authors, Journal of Nucleic Acids Research with the support of the “La Caixa” foundation identified the area on the surface of PDIA6 which binds to RNA molecules. With further research, this information can help design new therapy compounds that prevent the spread of melanoma from one part of the body to another. PDIA6 has previously been associated with the development of lymphoma, breast and lung cancer. If further research finds that PDIA6 also binds to RNA in other types of cancer, it can lead to therapeutic strategies that target the same action mechanisms in various tumors. This finding is possible thanks to the ‘RNA Fishing’ technique specifically designed to identify RNA binding proteins. This is a protein class that performs various biological functions, in cancer, provides resistance to cells by helping them adapt to changing environments and quickly respond to external threats, promoting cancer malignancy and resistance to therapy. This technique, also known as RNA Interactome Capture, involves fishing all RNA messenger in cells. The researchers then saw which protein attached to him, which informed the level and diversity of RNA binding activities that occur in the cell. “RNA binding protein is very interesting. Which protein might play an important role in the development of cancer, and conduct further experiments tell us how they work. We use this blueprint to uncover new vulnerability in melanoma, and we hope this can be repeated to find Target new therapy in other types of cancer as well, “explained Dr. Fátima Gebauer, a researcher at CRG and senior writer of this study. Of the hundreds of RNA binding proteins identified by this technique, researchers
I have Stage 3c melanoma skin cancer. I’ve had multiple surgeries, quarterly CT scans, needle biopsies and much more! This is an overview of everything I have gone through so far. I will have my 4th (of 12) treatment next week at Memorial Sloan Kettering. I am on Opdivo for 1 year. If you have any questions for you, leave it in the comments and I can get back to you. :-)—————————-Taken from Cancer.org August 2022 This explains exactly what I’ve been told by doctors and explains how it’s given to patients. Immunotherapy is the use of medicines to help a person’s own immune system recognize and destroy cancer cells more effectively. Several types of immunotherapy can be used to treat melanoma.Immune checkpoint inhibitors:An important part of the immune system is its ability to keep itself from attacking normal cells in the body. To do this, it uses “checkpoints,” which are proteins on immune cells that need to be turned on (or off) to start an immune response. Melanoma cells sometimes use these checkpoints to avoid being attacked by the immune system. But these drugs target the checkpoint proteins, helping to restore the immune response against melanoma cells.PD-1 inhibitors:Pembrolizumab (Keytruda) and nivolumab (Opdivo) are drugs that target PD-1, a protein on immune system cells called T cells that normally help keep these cells from attacking other cells in the body. By blocking PD-1, these drugs boost the immune response against melanoma cells. This can often shrink tumors and help people live longer. They can be used to treat melanomas:-That can’t be removed by surgery-That have spread to other parts of the body-After surgery (as adjuvant treatment) for certain stage II or III melanomas to try to lower the risk of the cancer coming backThese drugs are given as an intravenous (IV) infusion, typically every 2 to 6 weeks, depending on the drug and why it’s being given.
